Application note: Highly efficient method development using LC-MS for automated peak tracking

Peak tracking in liquid chromatography (LC) method development can be challenging due to retention variations and co-elution. Analytical conditions Table 1 presents the analytical conditions utilised to optimise the simultaneous analysis of six small molecular pharmaceuticals. Gradient elution was employed, using a 0.15 percent formic acid aqueous solution as the aqueous mobile phase and a combination of acetonitrile (ACN) and methanol (MeOH) as the organic mobile phase. The separation of individual compounds was thoroughly investigated by modifying the mobile phase composition of the organic solvent and adjusting the column oven temperature. Specifically, the ACN ratio in the organic mobile phase was adjusted from 0 percent to 100 percent in 10 percent increments (11 levels), while the column oven temperature was varied from 30°C to 40°C in 5°C increments (three levels).
