This webinar will appeal to all scientists and lab managers interested in the determination and quantitation of biomarkers and metabolites. The pressure to obtain high quality metabolism data, fast and confidently is ever increasing. Real world experience of the everyday challenges, and how cutting edge mass spectrometry is able to successfully overcome these challenges, will be presented by the two speakers.

The use of high speed high resolution MS combined with novel structurally intelligent software for the rapid identification, quantitation and correlation of specific metabolites and biomarkers across samples will be covered.

A question and answer session will follow the two talks. During which the speakers will be supported by a technical expert on the MS hardware and software from AB Sciex to address any questions the audience may have.

Keynote speakers

Dr. Axel Pähler, Senior Principal Scientist, Drug Metabolism and Pharmacokinetics, Hoffman La-Roche

Transforming data into knowledge: Use of HRMS instrumentation in Drug Discovery and Development

Dr Axel PaehlerDr. Pähler joined the Non-Clinical Drug Safety Department of F. Hoffmann-La Roche in 2001 where he applied his expertise in biochemical toxicology, drug metabolism and analytical chemistry to drug development projects. At Roche he then led the Drug Metabolism Group in support of drug metabolism studies for both Drug Discovery and Development until 2012. A strong focus of his work is on the development of new safety models to address and understand potential adverse side effects of new medicines. His research interests include biochemical processes in drug metabolism and disposition, mechanisms of reactive metabolite formation and toxicity, oxidative stress biomarkers as well as hyphenated mass spectrometry methods in metabolite and biomarker identification. Dr. Pähler currently holds a position as Senior Principal Scientist as Project Leader in DMPK at Roche in Basel, Switzerland.

Marieke Teppner, PhD Student, Drug Metabolism and Pharmacokinetics, Hoffman La-Roche

LC-MS/MS applications in biomarker research

Marieke TeppnerMarieke received her MSc in Pharmaceutical Sciences at the University of Kiel in 2009. She then carried out a practical training and master thesis in the DMPK Department of AstraZeneca plc. in 2010. Her work focused on the development of an assay for the determination of CYP1A2 and CYP2D6 contribution to hepatic clearance using human cryopreserved hepatocytes.

After graduation as pharmacist she started her PhD project in February 2011 in the Department of Non-Clinical Safety of Hoffmann-La Roche. Her work focuses on the characterization of risk factors for idiosyncratic liver toxicity via metabolic activation. Here, she investigates mainly oxidative stress biomarkers as danger signals for liver toxicity. A strong focus of her PhD project is on LC-MS/MS assay development and application to in vitro model systems. She also studies the correlation between biomarkers and reactive metabolite formation with the goal to link exposure with clinical outcomes.
